Protective Effects of Micro-Injected Adipose-Derived Mesenchymal Stem Cells on Ovarian Ischemia/Reperfusion Injury in Rats

Abstract
Introduction: The authors investigated the immunohistochemical, histological, and biochemical effects of adipose-derived stem cells (ADSC) which possess antioxidant and anti-inflammatory properties in the ovarian ischemia/ reperfusion rat model.
Materials and Methods: Rats were randomly allocated to sham, ADSC, torsion/detorsion, torsion/detorsion/ADSC groups. ADSCs were micro-injected to torsion/detorsion/ADSC during the detorsion procedure after a 6-h period of torsion. BrdU, caspase-3, TUNEL, eNOS, and iNOS staining were used. Advanced oxidation protein products (AOPPs), superoxide dismutase (SOD) activity, and levels of total sulfhydryl (TSH) were evaluated.
Results: Serum AOPP increased in the torsion/detorsion, tissue and serum AOPP levels in the torsion/detorsion/ ADSC group were decreased. Tissue SOD activity in torsion/detorsion and torsion/detorsion/ADSC groups was significantly decreased compared to the sham group. The iNOS was stronger in the torsion/detorsion group following torsion and detorsion but decreased after ADSC microinjection in the torsion/detorsion/ADSC group. ADSC treatment after torsion-detorsion also reduced caspase-3 immunoreactivity and apoptotic cell number.
Conclusion: ADSC implementation after torsion/detorsion might protect the ovary from ischemia/reperfusion injury.
